Plecanatide is a structural analog of human uroguanylin, and similarly to uroguanylin, plecanatide functions as a guanylate cyclase-C (GC-C) agonist. Both plecanatide and its active metabolite bind to GC-C and act locally on the luminal surface of the intestinal epithelium. Activation of GC-C results in an increase in both intracellular and extracellular concentrations of cyclic guanosine monophosphate (cGMP).
Plecanatide is used in adults to treat chronic idiopathic constipation (CIC; moeilijke of onregelmatige doorgang van ontlasting die aanhoudt 3 maanden of langer en niet wordt veroorzaakt door een ziekte of medicijn) en het prikkelbaredarmsyndroom met constipatie (IBS-C; een aandoening die maagpijn of krampen veroorzaakt, opgeblazen gevoel, en constipatie.)
Plecanatide zit in een klasse medicijnen die guanylaatcyclase-C-agonisten worden genoemd. Het werkt door de beweging van voedsel en afval door de maag en darmen te vergroten.
